I know tickets may be available at 7am on the day of a screening at the box office. How early should I line up?

If you have a Visa and don't mind the extra fee for online purchases ($5 vs. $2.25 in person, for entire transaction), I'd recommend crawling out of bed at 6:59am and hit up the website.
I've been to the box office in person at 7am a couple of times (because I was exchanging tickets/vouchers, which I can't do online), usually arriving at 6:30 am and being one of the first few in line. The big board showed my screening as available, but by the time I got to the head of the line, it was sold out.

I usually get there at 6 if at all possible, and that puts me usually in the first 40 or so people. If you're really worried, you will need to go really early to be in the first 10 or so people. But otherwise, 6-6:30 will set you up pretty well.

-7am same day tickets are only available at the venue box office of the screening you want tickets for and not anywhere else, is that right?

No, the festival box office sells tickets to all screenings. I'm not sure if they even do the 7 am tickets at the specific venue anymore.
